English: Homo- and heteropentameric glutamate-gated chloride channels (GluCl) are inhibitory neurotransmission mediators expressed in neural and muscle cells of nematodes, insects, and crustaceans. GluCl belong to the Cys-loop receptors family and consist of five subunits, which have the N-terminal extracellular ligand-binding domain (ten β-strains united in the immunoglobulin fold), the transmembrane domain of four α-helixes (M1-M4), and the intracellular domain. According to the family name, each subunit has the characteristic motif containing two cysteine residues connected with the disulfide bond (Cys-loops are purple in the illustration). An ionic channel pore is formed by M2 of each subunit; M1 and M3 separate M2 from a membrane lipid bilayer and participate in protein-ligand interactions.
Due to several binding sites, GluCl are targeted by many ligands divided into classes of positive and negative allosteric modulators based on their action. Ivermectin (colored in cyan), which is the widespread antiparasitic agent, belongs to the first class. Its binding site is located on the receptor transmembrane part between M3 of one subunit and M1 of an adjacent subunit. Ivermectin binding results in a cascade of conformational changes, which in the case of GluClα/β either leads to pore opening or enhances the ion current; in the case of GluClα (used in the illustration) it makes the receptor susceptible to glutamate and further activation by its binding. Consequently, ivermectin inhibits nematode motility, feeding, and reproduction leading to parasite death. The structure of C. elegans GluClα in complex with ivermectin (3.26 Å resolution) solved using X-ray crystallography by Hibbs and Gouaux (2011) was obtained from the RСSB PDB (ID: 3RHW) and visualized in the ChimeraX 1.4 by which images of three-dimensional complex model were made. Schemes of the cell membrane and the transmembrane part of GluCl were made and combined with images of the three-dimensional complex model in Adobe Illustrator. Structural and functional aspects of ivermectin interactions with known macromolecules can be used during a search for ivermectin binding sites on new targets.Українська: Гомо- та гетеропентамерні глутамат-залежні хлоридні канали (GluCl) є посередниками у передачі інгібуючих імпульсів, що експресуються у нервових та м’язових клітинах нематод, комах та ракоподібних. GluCl належать до родини Cys-петльових рецепторів та складаються з п’яти субодиниць, кожна з яких містить: N-кінцевий позаклітинний ліганд-зв’язуючий домен (десять β-тяжів, об’єднаних в імуноглобулінову складку); трансмембранний домен, утворений з чотирьох α-спіралей (M1-M4); внутрішньоклітинний домен. Відповідно до назви родини, субодиниці рецептора мають характерний мотив, що містить два цистеїнові залишки, об’єднані дисульфідним зв’язком (на зображенні Cys-петлі позначені пурпурним), та сполучає позаклітинний та трансмембранний домени. Пора іонного каналу формується M2 кожної субодиниці; M1 та M3 відділяють M2 від оточуючого ліпідного шару та беруть участь у взаємодії з лігандами.
Наявність декількох сайтів зв’язування роблять GluCl ціллю для багатьох лігандів, які за способом дії поділяються на класи позитивних та негативних алостеричних модуляторів. До перших, зокрема, належить й поширений антипаразитичний агент івермектин (позначений ціановим). Сайт зв’язування івермектину з GluCl знаходиться у трансмембранній частині рецептора між M3 одної субодиниці та M1 прилеглої субодиниці. У результаті зв’язування виникає каскад конформаційних змін, що у випадку GluClα/β призводять до відкриття пори або посилення вже існуючого потоку йонів, а у випадку GluClα (використаний в ілюстрації) роблять рецептор чутливим до глутамату та подальшої активації при зв’язуванні з останнім. У результаті дії івермектину нематода втрачає здатність до руху, живлення та розмноження, що призводить до смерті паразита. Структура GluClα C. elegans у комплексі з івермектином з роздільною здатністю 3.26 Å, вирішена через рентгеноструктурний аналіз (Hibbs and Gouaux 2011), була видобута з RСSB PDB (ID: 3RHW) та візуалізована у програмі ChimeraX 1.4, за допомогою якої були отримані зображення тривимірної моделі комплексу та івермектину. Cхематичні зображення мембрани та трансмембранної частини GluCl були зроблені та об’єднані з тривимірними зображеннями у Adobe Illustrator.
